Lead Investigator: Guojun Bu Institution : Mayo Clinic E-Mail : Bu.Guojun@mayo.edu Proposal ID : 485 Proposal Description: Numerous studies demonstrated that APOE4 accelerate brain volume loss in especially AD-vulnerable areas, such as hippocampus and temporal cortex even without development of Alzheimer???s disease. On the other hand, a few study investigated the effect of APOE2, protective risk factor for AD, on brain volume change. We aims to compare the effects of APOE2, APOE3, and APOE4 genotype on brain volume in different brain area of people with/without cognitive decline by cross sectional and longitudinal approaches. Specific aim 1: We will longitudinally study how the presence of AD-risk factor APOE4 and AD-protective factor APOE2 affect the brain volume change in people with cognitively normal, MCI or Alzheimer???s disease patients. Specific aim 2: We will cross-sectionally study how the presence of AD-risk factor APOE4 and AD-protective factor APOE2 affect the brain volume in people with cognitively normal, MCI or Alzheimer???s disease patients.
